Alkaline phosphatase is assayed at a concentration of 1 ug/ml at room temperature in a 0.1 M glycine buffer, pH 9.5 in the presence of 5mM MgCl2 as a co-factor in the reaction. The substrate 4-nitrophenol phosphate (4-NPP) is added in a concentration range of between 0.05 mM and 3.0 mM. The appearance of the product, 4-nitrophenol (4-NP) can be monitored at 420nm (E= 15mM -1cm -1).
