I know it's kinda predictable but my favourite novels really are To Kill A Mockingbird by Harper Lee and Pride and Prejudice by Jane Austen.

That said, Holistic Detective Agency is great.

Actual post content: The Illium/Olympos books by Dan Simmons are a great read, if you like bizarre science fiction involving pseudo-Greek Gods, irreverent technobabble, and an obscenely powerful side-character getting crippled using a dash of trickery and chemistry.

I find it really hard to choose only one book. I've read too many (mostly sci-fi/fantasy), and I tend to fall in love with whatever it is I'm reading at the time. :p

Probably the Birth of Tragedy by Nietzsche. This had a profound effect on me when I first read it, even though I only understood half of it (knowing nothing about the Ancient Greeks).
